What is unusual about the residues in the chain?
Several of the thirty-nine are non-proteinogenic, chosen to block enzymatic cleavage sites a natural sequence would present, and one lysine side chain additionally carries a C20 diacid through a linker. None of those positions can be assembled from a standard amino-acid set, which is part of why long engineered peptides are harder to source to a consistent specification.
What should the certificate cover for a fill this large?
The same fields as any lot — purity by HPLC with the method conditions stated, identity by mass, and the lot number — but they warrant closer reading, because a 40mg vial is likely to be the source for an entire assay series rather than a single run. One lot carrying a whole programme is only an advantage if its documentation is sound.
Why can a lipidated peptide behave awkwardly in aqueous buffer?
Attaching a long diacid chain to a polar peptide makes the molecule amphiphilic, and amphiphiles can form micelles or adsorb to vessel surfaces at concentrations where an ordinary peptide would simply dissolve. Quantitative work should allow for that; establishing suitable conditions is the researcher's call rather than ours.
