What does the research on NAD+ investigate?
NAD+ is not a peptide. It is a dinucleotide - two nucleotide units joined through a pyrophosphate linkage - and it is best understood as a coenzyme, the molecule that carries electrons in a large family of oxidation and reduction reactions. That role is why it is found in essentially every living system, and it is also why the interesting analytical question is not whether the material is pure but which of its two states it is in. NAD+ is the oxidised form and accepts a hydride ion to become NADH, the reduced form, and the two are distinct molecules with different absorbance, different behaviour on a column and a small but measurable mass difference. Work with this material is generally work on that redox couple. The pyrophosphate bond at the centre of the molecule is also its weak point, since it hydrolyses under conditions that a peptide would tolerate. Nothing here describes a use, benefit, dose or effect in people.
Is NAD+ legal in the UK?
NAD+ is not a licensed medicine in the United Kingdom and is not listed in any schedule of the Misuse of Drugs Act 1971. Selling it as a research chemical is a lawful trade; what the law restricts is the purpose it is sold for, not the fact that it is sold.
Three pieces of legislation frame that distinction. The Human Medicines Regulations 2012 make it an offence to sell an unlicensed product for human use. How are identity and quality assured?

Purity is measured by reversed-phase HPLC at an independent laboratory. Synthesis and supply

Why is the 340nm band useful?
NAD+ itself does not absorb appreciably at 340nm; its reduced form NADH does. Following that band rise or fall is the standard way to track a dehydrogenase reaction in real time, and it is the reason so many coupled assays are built around this cofactor.
How stable is the material in solution?
Much less stable than the dry solid. Aqueous NAD+ degrades over time and does so faster as pH rises above neutral and as temperature increases, so solutions are normally made up for the run in hand rather than stored. Any shelf figure quoted on the lot documentation applies to the dry powder, not to a prepared solution.
Is it supplied as the free acid or a sodium salt?
Both forms circulate in the reagent market and they carry different formula masses, so this is worth confirming before a weighed mass is converted into a concentration. The lot documentation states the form that was tested.

How should NAD+ be stored?
In the form supplied, as a lyophilised powder, the material is stable kept cold, dry and dark. For powder, that means a freezer at around minus 20 degrees Celsius, desiccated, with the vial sealed and shielded from light, which is how it should stay until you are ready to work with it. Bringing a vial to room temperature before opening avoids condensation on the contents. Once in solution the material is less stable than the powder and should be kept cold and used within a short working window. Choosing a solvent and a storage regime for your own work is your call, not ours.
Do I need bacteriostatic water to prepare it?
A lyophilised powder needs a solvent added before it can be worked with. Bacteriostatic water, which is water with 0.9% benzyl alcohol, is the common choice because the preservative extends the working life of the solution once the vial has been opened, and we supply it in 3ml and 10ml vials. What is the difference between NAD+ and NADH?
Two electrons and a proton. NAD+ is the oxidised form and accepts a hydride ion to become NADH, the reduced form. They are chemically distinct species with different ultraviolet absorbance - which is convenient, because it means the ratio between them can be followed directly by spectrophotometry - and different retention behaviour on a separation method. When a certificate describes the purity of this material, knowing which of the two states is being described is essential.
Why does the pyrophosphate bond matter?
It is the most labile part of the molecule and the first thing to fail. Two phosphate groups are joined through an anhydride linkage, and that bond hydrolyses in water under conditions a peptide backbone would comfortably survive, particularly at extremes of pH or elevated temperature. Because the products of that hydrolysis are themselves nucleotides, they can be mistaken for impurities of manufacture rather than products of storage.
Can NAD+ be analysed by reversed-phase HPLC?
Not usefully on its own. The molecule carries two phosphate groups and is strongly polar and negatively charged at neutral pH, so it has almost no interaction with a reversed-phase stationary phase and elutes essentially with the solvent front. Methods for this material use ion exchange, ion pairing or a specialised column chemistry. A generic peptide gradient will not separate it from anything.
Why does the material take up water?
Because it is hygroscopic and readily absorbs moisture from the air. That is a genuine practical concern for a compound sold by weight, since material that has taken up water weighs more than the material itself, and a fill verified by weighing alone can be misleading. It is another reason to keep the vial sealed and to bring it to room temperature before opening, so that condensation does not form on the contents.
