What does the research on Melanotan-1 investigate?

Melanotan-1, also known as afamelanotide, is a synthetic thirteen-residue analogue of alpha-melanocyte-stimulating hormone, and its sequence is a lesson in designing around a known weakness. The parent hormone carries a methionine that oxidises readily; this analogue replaces it with norleucine, a non-natural residue of almost identical shape and size that cannot oxidise, so the molecule keeps the geometry and loses the liability. The chain also carries a D-configured phenylalanine and is amidated at the C-terminal end, both of which make it harder for enzymes to take apart. Analytically this is a linear peptide with one remaining tryptophan, so oxidation is still the change to watch for, but the main modification is deliberate rather than a defect. The published research uses the sequence in melanocortin receptor binding studies. Nothing here describes a use, benefit, dose or effect in people.

What do the Nle4 and D-Phe7 substitutions change?

Norleucine at position four removes the oxidation-sensitive methionine side chain, replacing the thioether with an unreactive hydrocarbon of similar length. D-phenylalanine at position seven inverts one stereocentre, which alters the local backbone geometry and makes the chain a poorer substrate for L-specific proteases. Both are structural features written into the sequence itself and appear as such on the certificate.

Why are both termini capped?

The amino end carries an acetyl group and the carboxyl end an amide, so neither presents a free charged group. Analytically that means the observed mass differs from the uncapped sequence, and the peptide offers no free alpha-amino group for chemistries that require one.

How should MT-1 / Melanotan-1 be stored?

Why does the analogue contain norleucine?

To remove a point of instability. Norleucine is a non-natural residue with a hydrocarbon side chain the same length as methionine's but without the sulfur, which is the part that oxidises. Swapping one for the other leaves the shape of the molecule essentially unchanged while removing the chemistry that would have degraded it. It is a good illustration of how a single residue substitution can be made for stability rather than for function.

How does this differ from Melanotan-2?

They are different molecules with different lengths and different structures: this one is a linear thirteen-residue analogue, while Melanotan-2 is a cyclic analogue of ten residues formed from a different parent sequence. Their molecular weights differ, their retention behaviour differs and their handling differs. They should not be treated as two strengths of the same material, because they are not the same material.

What is the relationship to alpha-melanocyte-stimulating hormone?

This is a synthetic analogue of that hormone, meaning the sequence has been altered rather than extracted, and the changes are the point of it. The relationship matters for understanding why the molecule is structured the way it is, but it does not make the two interchangeable and it describes nothing about what the material is for.

Where is the remaining oxidation risk?

At the single tryptophan residue. Removing methionine closed one route but tryptophan remains, and it oxidises readily, adding sixteen daltons and shifting retention. On a purity trace it appears as a closely related second peak. Keeping the vial sealed, cold and dark, and minimising how often it is opened, is the practical mitigation.
