Glutathione in research
Glutathione is held together by an unusual bond. Its N-terminal glutamate is linked through the side-chain carboxyl rather than the alpha-carboxyl, giving a gamma-glutamyl amide that ordinary peptidases cannot cleave — which is why gamma-Glu-Cys-Gly survives where a conventional Glu-Cys-Gly would not. The reactive centre is the cysteine thiol, and it is also the handling problem: exposed to air or standing in neutral buffer, reduced glutathione (GSH) oxidises to the disulfide dimer GSSG, a separate species at roughly twice the mass. A purity figure on this material therefore means little until the certificate says which form was measured. How are GSH and GSSG distinguished on a certificate?
GSH is reduced glutathione, the monomer with a free cysteine thiol. GSSG is the oxidised dimer, two molecules joined by a disulfide bridge at roughly twice the mass. They are separate substances with separate registry numbers, and a purity figure is only interpretable once the certificate states which was measured.
Why is the glutamate linkage described as gamma?
The amide bond runs from the side-chain carboxyl of glutamate to the amino group of cysteine, not from the alpha-carboxyl as in a normal peptide bond. Standard peptidases do not recognise that linkage. It is a defining structural feature of the molecule rather than a modification made to it.
What makes the dry solid the stable form?
The free thiol oxidises readily in solution, and trace metal ions accelerate it. As a sealed, dry, frozen solid that route is largely closed off. Solutions of the reduced form are normally prepared for the work in hand rather than held.
